党参多糖对环磷酰胺所致小鼠贫血的治疗效果观察

摘要:

目的探讨党参多糖对环磷酰胺(CTX)所致小鼠贫血的疗效。方法将60只SPF级健康雄性BALB/c小鼠均分为空白组、模型组和低、中、高剂量组及阳性对照组6组,除空白组外,其他5组分别于实验第1天按100 mg/kg剂量腹腔注射CTX,每日1次,空白组予等容积生理盐水腹腔注射,连续7 d。实验第2天注射CTX后2 h,低、中、高剂量组分别予党参多糖10、20、 40 ml/kg灌胃,阳性对照组予生血宁溶液400 mg/kg每日1次灌胃,连续7 d。采用全自动血球计数仪测定各组红细胞(RBC)、血红蛋白(Hb)及红细胞压积(HCT),采用ELISA法测定促红细胞生成素(EPO)、多能集落刺激因子(multi-CSF)水平,采用流式细胞仪测定外周血CD71+和Ter119+水平。结果与空白组比较,模型组外周血RBC、Hb和HCT均显著降低(P<0.05);与模型组比较,低、中、高剂量组RBC、Hb和HCT均显著升高(P<0.05);低剂量组、中剂量组RBC、Hb、HCT明显低于阳性对照组(P<0.01)。与空白组比较,模型组外周血multi-CSF和EPO水平均显著降低(P<0.05);与模型组比较,低、中、高剂量组multi-CSF和EPO水平均显著升高(P<0.05);低、中、高剂量组multi-CSF均低于阳性对照组(P<0.01),且随着干预剂量的增加multi-CSF呈升高趋势;低、中剂量组EPO明显低于阳性对照组(P<0.01)。模型组CD71+和Ter119+水平显著低于空白组(P<0.01);与模型组比较,低、中、高剂量组CD71+和Ter119+水平均显著升高(P<0.01);低、中剂量组CD71+和Ter119+水平均低于阳性对照组(P<0.01)。结论党参多糖能有效改善CTX所致小鼠贫血。

Abstract:

Objective To investigate the therapeutic effectiveness of Codonopsis Pilosula Polysaccharide on mice with anemia induced by Cyclophosphamide(CTX). Methods A total of 60 SPF healthy male BALB/c mice were equally divided into 6 groups blank group, model group, low, medium and high dose groups and positive control group. Except for blank group, the other 5 groups were intraperitoneally injected with CTX by the dose of 100 mg/kg on the first day of the experiment for once a day. The blank group was intraperitoneally injected with the equal volume of physiological saline for 7 d continuously. At 2 h after CTX injection on the second day of the experiment beginning, the low, medium and high dose groups were given 10, 20 and 40 ml/kg Codonopsis Pilosula Polysaccharide by gavage respectively, while positive control group was given 400 mg/kg Shengxuening Solution by gavage once a day for 7 d continuously. Levels of red blood cells(RBC), hemoglobin(Hb) and hematocrit(HCT) were detected by automatic blood cell counter, and levels of erythropoietin(EPO) and multi colony stimulating factor(multi-CSF) were detected by enzyme linked immunosorbent assay(ELISA), and levels of CD71+ and Ter119+ in peripheral blood were detected by flow cytometry in each group. Results RBC, Hb and HCT levels in peripheral blood in model group were significantly lower than those in blank group(P<0.05). RBC, Hb and HCT levels in low, medium and high dose groups were significantly increased compared with those in model group(P<0.05); RBC, Hb and HCT levels in low and medium dose groups were significantly lower than those in positive control group(P<0.01). Levels of multi-CSF and EPO in peripheral blood in model group were significantly lower than those in blank group(P<0.05). Levels of multi-CSF and EPO in low, medium and high dose groups were significantly increased compared with those in model group(P<0.05). The multi-CSF levels in low, medium and high dose groups were significantly lower than that in positive control group(P<0.01), and multi-CSF level was increased with increasing intervention dose. EPO levels in low and medium dose groups were significantly lower than that in positive control group(P<0.01). Levels of CD71+ and Ter119+ in model group were significantly lower than those in blank group(P<0.01). Levels of CD71+ and Ter119+ in low, medium and high dose groups were significantly increased compared with those in model group(P<0.01). Levels of CD71+ and Ter119+ in low and medium dose groups were significantly lower than those in positive control group(P<0.01). Conclusion Codonopsis Pilosula Polysaccharide may effectively improve mice with anemia induced by CTX.
